RNA−protein interactions are important interactions among biological molecules as well as protein−protein interactions. The identification of the counter part RNA of proteins has been considered to be valuable to understand the roles of RNA molecules and discover new therapeutic targets. Cross-linking methods are powerful tools for identification of the interaction partner of biological molecules of interests, since a covalent bond is formed allowing its detection under stringent conditions. Recently, photo-activable diazirine has been used as a photo−cross−linking method, which is advantageous over chemical cross−linking methods. PhotoMet, a photo-activable amino acid bearing diazirine, was incorporated into a library of amphiphillic peptides. Each peptide in the library was subjected to be photo-crosslinked with known RNA targets including RRE RNA. Some of theses peptides formed the covalent adduct in position selective manners, suggesting that the incorporation of photoMet to RNA binding peptides could be a promising research tool for the study on RNA-protein interactions.
