Design and Synthetic Studies of Novel Guanidinium-Rich Molecular Transporters Using scyllo-Inositol as the Scaffold

For the effective deliver to the desired site, a drug molecule should pass through biological barriers, such as cellular membrane, blood-brain barrier (BBB). Certain guanidinium-rich transporters (GRTs) have been found to readily overcome those barriers. Previously we prepared GRTs based on inositol-dimer scaffolds carrying 8 guanidines linked by ω-aminocarboxylates with variable chain length. They showed good cellular uptake and were distributed in the liver, kidney, especially brain. However, the synthesis of inositol-dimer based molecular transporters is cumbersome and inefficient. Therefore, we designed and synthesized scyllo-inositol based GRTs containing branched linkers and examined their cellular uptake, localization in cells, and tissue distribution in mouse.
