It is well-known that the programmed -1 ribosomal frameshifting (-1RF) regulates synthesis of proteins. Many viruses including RNA viruses use the programmed -1RF mechanism to produce more than one polypetide from single mRNA. Two RNA cis-elements - the slippery sequence and a RNA structural element, mostly RNA pseudoknots - are prerequisite for efficient -1RF. In particular, thermodynamic stability and/or kinetic control of RNA pseudoknot folding/unfolding process are important in regulating the efficiency of -1RF. Thus, it is possible that changing the stability of RNA pseudoknot by small ligands can alter -1RF efficiency. In this study we synthesized H4, which screened from the previous work (Park et al., 2008), that affect the -1RF by interacting with the biotin-binding RNA pseudoknot. A synthesis of H4 was achieved in four steps from L-(+)-lysine as a starting material. We plan to examine the effect of H4 on the efficiency of -1RF system in vitro.
