Biodegradable polymers have been extensively investigated because of regulating drug release rate easily, obviating the need to remove the device, and good biocompatibilty. In this study, Highly porous biodegradable poly(D,L-lactic-co-glycolic acid) (PLGA) microspheres were fabricated by a single water-in-oil emulsion solvent evaporation technique using Pluronic F127 as an extractable porogen and lysozyme as a model protein was incorporated into porous microspheres by a simple solution dipping method. For their controlled release, porous microspheres containing protein were treated with water-miscible solvents in aqueous phase for production of pore-closed microspheres. The morphology and drug distribution of the protein loaded microspheres were investigated by scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM), respectively. Moreover, structural integrity of released protein after the treatment was observed by enzymatic activity assay, size-exclusion chromatography (SEC), gel electrophoresis (SDS-PAGE.). Also,the structure of released Lysozyme was determined by Fourier transform infrared (FTIR).
