Preconcentration for Proteomic Sample Using Free-Flow Isoelectric Focusing in PDMS Microfluicid Device

For the separation of various proteins in a micro-liter volume, microfludic free flow isoelectric focusing (μ-FFIEF) technique is used. For the prefractionation and preconcentration of proteins mixture, palladium electrodes are patterned on the glass substrate and coated with TEOS-sol to enhance the bonding with PDMS. The microfluidic channel is made of PDMS by photolithography molding technique. In this study, for the elimination of electroosmotic flow, dynamic coating of SiO2 over PDMS microcannel is used. The device was tested by adding pH indicator in the MES buffer solution used in actual experiment. Here, the optimum pH, buffer concentration and applied electric voltage were found as 6.0, 1 mM and 1.5Volts, respectively. Self-illuminating proteins, RFP and EGFP, were chosen as model proteins. Under optimum conditions, RFP and EGFP, were successfully concentrated at around 214 μm and 357 μm from anode, respectively. The time required for full concentration in the 0.5 mm channel was about 120s, longer than the focusing time in the channel without glass coating which is approximately 30 s. But, the break-down of electrode by electrolysis started immediately when electric field is applied to the device without glass coating.
