Glucose Bioprobes : their synthesis and biological applications

In the previous research, we reported that Cy3-Glc-α (named GB1-Cy3) is a novel bioprobe for bioimaging and screening for new anti-cancer agents through the efficient in-vitro monitoring of glucose trafficking. As a extension of our study, we designed and synthesized on array of novel fluorescent glucose bioprobes(GB) with the variation of linkers and fluorescent dyes. We monitored the specific uptake of GB by fluorescent microscope for bioimaging and Fluorescent Cell sorting (FACS) for quantification analysis. High concentration of D-glucose in medium can effectively reduce the cellular uptake of GBs and the significant enhancement of GB was consistently observed in the D-glucose depleted medium. In comparison the high concentration of L-glucose has no effect on cellular uptake of GBs, which confirms the specific cellular uptake of GB through GLUT (glucose transporter). We are particularly interested in GLUT4, which is actively regulated its translocation in adipose and muscle. With our novel array of GBs, we pursued the systematic studies regulated glucose uptake in adipose and muscle cells under the treatment of Insulin, Rosiglitazone or AICAR. We believe that these glucose bioprobes can provide powerful research tool to study Type 2 Diabetes and Obesity via the application in HTS (High-Throughput Screening) and Bioimaging.
