Reversible phosphorylation of protein tyrosine residues is a unique process, mediating cellular signal transduction, gene regulation, cell mobility, and so on. Human protein tyrosine phosphatase (PTP) family, consisting of 107 members, catalyzes a hydrolytic dephosphorylation of phosphotyrosine. Several PTPs have been being therapeutic targets for human diseases such as cancer, diabetes, inflammation, obesity, etc. Selective PTP inhibitors thus are expected to be therapeutics as well as potential chemical tools for functional study of the corresponding PTPs. To find leads for selective PTP inhibitors, we have successfully overexpressed and purified 77 human PTPs from E. coli systems, which were applied to screen a chemical library in a high-throughput manner. We have also developed novel fluorescent probes, which can be applicable to profile cellular PTP activities.
