We present a facile method of achieving low surface coverage of gold nanoparticles (AuNPs) on indium tin oxide (ITO) electrodes which afford both low backgrounds current and good electrocatalytic properties for sensitive electrochemical detection. The AuNPs are formed on the amine-functionalized ITO surface by electrostatic adsorption of AuCl4− followed by chemical reduction. The AuNPs formed in this way provide efficient sites for avidin immobilization. To fabricate immunosensor, biotinylated antimouse IgG is specifically bound to avidin immobilized on AuNPs. Subsequently, target mouse IgG is captured, and then alkaline phosphatase-conjugated antimouse IgG is bound. Alkaline phosphatase catalyses the conversion of 4-aminophenylphosphate to 4-aminophenol, and then the amplified 4-aminophenol is electrooxidized on the AuNPs to obtain an electrochemical signal. The detection limit of the sensor is 10 pg/mL mouse IgG.
