An antisense oligonucleotide (ASO) selectively binds to a target mRNA and inhibits the synthesis of the protein encoded by the mRNA if properly delivered into cell. In this regard, ASOs can be developed into drugs for diseases, which would be practically impossible to treat with currently popular approaches such as small molecule drugs, monoclonal antibodies, protein drugs, and so on. Unfortunately most ASOs including siRNA rarely penetrate cell membrane and would be difficult to show therapeutic effects unless properly delivered to target tissues. Mipomersen is an ASO drug candidate in late phase III clinical trials and preferentially distributes to the liver for its anti-aetherosclerosis activity. Mipomersen is expected to be essentially the first ASO drug reaching the market. In this presentation, will be discussed critical issues for drug deveolpment of ASOs with emphasis on the cell penetration of oligonucleotides.
