Biochemistry textbooks state a golden rule in protein science; a protein must have a uniquely and well-defined three-dimensional structure to exhibit proper function. On the other hand there have been at least dozens of cases where one could not assign a unique structure to a protein. Bioinformatics predictions and NMR experimental studies have demonstrated that there are many proteins named “intrinsically unfolded/unstructured proteins (IUPs)” which do not obey the golden rule of structural biology and do not have globular tertiary structures, yet are fully functional. Some believe that fraction of IUPs in the whole protein kingdom may reach 50% or more. It is not clear if such proteins have found a place in the mainstream of protein science, nevertheless it is obvious that protein chemists should look into these peculiar proteins before they aspire to get a complete picture of protein interaction network. This talk will present a critical view on IUPs.
