Noninvasive imaging technique is essential for the diagnosis and monitoring in biomedical application. But each of single imaging techniques has limitations. Therefore, the combination of these techniques was necessary to complement the limitations. In this report, we developed radiolabeled NIR-fluorescent magnetic silica nanoparticles, 68Ga-(MNP-SiO2(NIR797)) for PET/MR/NIRF images. This advanced nanoprobe shows strong contrast effect for PET/MR/NIRF images, biocompatibility for biomedical application, and the enhancement of the fluorescent intensity and stability compared to the free dyes. We also demonstrated our probe’s contrast effect in vivo through sentinel lymph node (SLN) mapping by using PET/MR/NIRF images. Moreover, to evaluate the enhanced NIRF property by encapsulation of NIR fluorescent dye in the silica shell, we applied our probe for cell trafficking in the deep tissue (brain, spinal cord and liver). We injected cells labeled our probe into the deep tissue and then we obtained well distinguished NIRF images in the injected area. In conclusion, we developed radiolabeled magnetic silica nanoparticles that encapsulate NIRF dye in the silica to decrease toxicity, enhance the fluorescence stability and deep tissue imaging. We anticipate that this advanced probe provided a complementary role in imaging diagnostics and cell trafficking.
