Protein tyrosine phosphatase (PTP) family catalyzes a hydrolytic dephosphorylation of phosphotyrosine, which affects various cellular signaling and disease status. High similarities in the structure and catalytic mechanism between PTP family members have hampered developing selective inhibitors for specific PTPs of medicinal interests. Considering the structural similarities of the PTPs envisioned that their selective inhibitors could be derived from even a single structural scaffold. This idea led us to search structural scaffolds, which are able to generate diverse chemical structures of potential PTP inhibitors by a robust chemistry. Triazole acetic acid (I) is one of the desired scaffolds. The design, synthesis, and inhibitory activity of a series of compounds related with I will be presented.
