Currently, many biologists have been researching about the Mitochondria having a critical role in celluar respiration. Mitochondria are power sources of the cell because their primary function is to convert organic materials into energy in the form of ATP which is an energy molecule. Mitochondria have their own DNA and usually they are rod-shaped, ranging from 1 to 4µm in length. Flow Field Flow Fractionation (FlFFF) is known as a universal technique for the separation and characterization of proteins, polymers, colloid particles, macro-molecules and organelles. In this study, we characterized the mitochondria from the whole cell extracted from rat liver by using an Asymmetrical FlFFF (AFlFFF) and scanning electron microscope (SEM). Finally, each fraction was analyzed secondarily for proteome analysis by using reverse phase liquid chromatography-tandem mass spectrometry (RPLC/MS/MS) for exploring mitochondrial proteins.
