Glutamate-induced HT22 cell death is distinct from classical cell death as it is mainly caused by the oxidative stress resulting from depleting GSH via that extracellular glutamate inhibits cystine uptake through cystine/glutamate antiporter. The oxidative stress results in oxidative products of lipids, proteins, and DNA, which are frequently associated with neuropathological conditions. In our screening system, a natural product, previously known as a neuroprotectant and/or anticancer, showed a protective activity of HT22 cell from glutamate-induced cytotoxicity. An affinity column made of the compound could separate high affinity proteins from HT22 cell lysate, and the proteins have been frequently found as oxidatively damaged forms in the neuronal cells of individuals with various neurodegenerative diseases. The details will be presented.
