Microscale device fabrication and microfluidics have enabled the miniaturization of chemical and biological analysis, which is motivated by the need to reduce the use of expensive reagents and precious samples, and to increase the analysis speed. In addition, the microfluidic system is applied with a promise of conducting a complete assay on a single chip. We have utilized a microfluidic instrument, comprised of a hydrodynamic flow controller, optical system, and a plate handling system, to develop a high-throughput screening system for enzymes. Employing a microfluidic chip fabricated on glass, fluorogenic assays for protease and phosphatases were performed. The system was then optimized for high-throughput screening and validated by use of inhibitory compounds.
