Solid-phase synthesis has emerged as a powerful technique in generating combinatorial libraries of small organic molecules useful for drug discovery. Previously, we reported the solid-phase synthesis of N,N-disubstituted hydroxylamine and hydroxypiperazine derivatives based on the oxidation-Cope elimination of polymer-bound phenethylamine linker with m-CPBA. The desired compounds could be obtained in high purities and yields without any significant contamination at mild condition. In our research program for the development of various benzopyran derivatives, we would like to report the synthesis of spiro-benzopyran derivatives containing hydroxylamine and tertiary amine moiety using Cope-elimination and Hofmann-elimination cleavage of polymer-bound phenethylamine linker.
