ECL-Biosensors Based on Carbon Nanotube/Titania-Nafion Composite Films

An ethanol biosensor based on electrogenerated chemiluminescence detection has been investigated. The Ru(bpy)32+ ECL sensor can detect NADH with good sensitivity, but not detect NAD+. Moreover, the titania-Nafion composite are biocompatible and carbon nanotube (CNT) with mechanical rigidity, electrical conductivity, and chemical stability can be dispersed in the titania-Nafion composite. Therefore, the present Ru(bpy)32+ ECL sensor based on the MWCNT-titania-Nafion composite films can be easily modified into dehydrogenase-based ECL biosensors simply by co-immobilizing NADH-producing dehydrogenase enzymes such as alcohol dehydrogenase (ADH). The NADH produced from the enzymatic reaction is detected with the Ru(bpy)32+ ECL sensor. Therefore, ECL biosensor based on the MWCNT-titania-Nafion composite films has been studied for the determination of ethanol. Under the optimum conditions (ADH unit, NAD+ concentration, pH, and working potential), calibration curve for ethanol has been constructed with the ECL biosensor. The present biosensor shows good sensitivity and responds to primary alcohols such as n-propanol and n-butanol with ECL responses of about 10% and 5%, respectively, relative to that for ethanol. In contrast, the methanol was not detected with the present biosensor.
