 Behcet's disease (BD) is a multi-system inflammatory disorder of unknown etiology. Inflammation in the disease is thought to be mediated by cytokines derived from T-helper type 1 (Th1) lymphocytes. This study aimed at investigating the associations between Behcet's disease in Korean people and gene polymorphisms. This study tries to determine whether NRAMP1, IL-18 and IL-6 polymorphisms are associated with susceptibility to Behcet's disease in Koreans. The three NRAMP1 polymorphisms (A318V, D543N, (GT)n microsatellites in promoter region), two IL-18 polymorphisms ( -607 C/A and -137 G/C ) and two IL-6 polymorphisms (-174G/C, VNTR in the 3'flanking region) were typed by SS-PCR (Sequence-Specific Polymerase Chain Reaction), VNTR(Variable Number of Tandem Repeat), RFLP(Restriction Fragment Length Polymorphism), DHPLC (Denaturing High Performance Liquid Chromatography) and Sequencing. There were no evidences for genetic association conferred by the IL-6prom polymorphism. However, significant differences in the IL-6vntr genotype and allele frequencies were found between patients with BD and controls.
