Determining the phosphorylation site of proteins by kinases is essential for the study of regulation of cell functions. Therefore a number of methods have been studied to identify the potential kinase substrates by combinatorial synthesis of peptide libraries. We have developed the one-bead one-compound (OBOC) combinatorial library method that is efficient for the screening the substrate specificity. In this way, each of the resin beads carries not only the full-length product but also small amounts of sequence-specific N-terminated products. The sequence of the full-length peptide is determined by using MALDI-TOF MS, which is used to measure mass differences between the adjacent members of the N-terminated products. We also introduced mass sensitizer to increase the sensitivity in analyzing the peptide sequence.
