An RNA Aptamer Can Discriminate a Specific Conformation of Calsenilin Protein

Multiple functions of a protein can be derived from many conformations of a certain protein. Therefore, it is necessary to develop antibody-like molecules to elucidate conformational-specific functions of the protein. We now apply RNA aptamers and the SELEX (Systematic Evolution of Ligands by EXponential enrichment) technology to discriminate many conformations of calsenilin. Calsenilin protein is involved in Calcium-mediated apoptotic signaling and the production of Amyloid-beta 42. It is also known that the conformation of the protein is quite different in the presence and absence of calcium. We selected out RNA aptamers that could only recognize calcium-bound conformation of the protein. RNA pull-down and surface plasmon resonance assays showed that the specific RNA clone discriminates calcium-rich conformation from calcium-deficient one. RNA foot-printing assay showed that the binding site of the RNA aptamer is a loop region that can discriminate each conformational state of calsenilin. Development of intramer system to test the modulatory effect on calsenilin protein in neuronal cells is in progress.
