New Heteroconjugates for 16S Ribosomal RNA Decoding Region; A possible New Antibiotics

While a development of completely new antibiotics may not an easy task, an improvement of specificity by tethering two known drugs is a relatively easy task. A strategy involves the addition of a loop-specific antibiotic to a stem-specific aminoglycoside to make heteroconjugates. These conjugate molecules have at least two different sites to interact with stem-loop RNA motifs, affording specific affinities against RNA targets. We recently designed and synthesized the heteroconjugates and applied these molecules to RRE and TAR RNA successfully. This time, we are applying these heteroconjugates to 16S A-site rRNA decoding region as a new RNA target using a fluorescence anisotropy technique. More than 10 times of affinity enhancement was observed in a selected heteroconjugate comparing to that of neomycin, suggesting that this type of compounds could be a new potential antibiotic targeting 16S a-site rRNA.
