Autoantibodies are frequently found in many autoimmune patients. One of these autoantibodies are anti-DNA antibodies, which can recognize DNA, and can cleave DNA as well. In order to investigate catalytic activity of these autoantibodies, monoclonal antibodies has been raised in our laboratory. Among these monoclonal antibodies, antibody (Ab) G1-5 showed the strongest catalytic activity for the initial screening with calf thymus DNA. In this study, we further investigates kinds of catalytic activities that antibody Ab G1-5 possess using a various standard substrates including phosphatase and glycosidase activities. Enzymatic constants of Ab G1-5 were determined and were compared with those of other catalytic antibodies. Phosphatase and glycosidase activities of Ab G1-5 were totally inhibited by DNA, suggesting the catalytic activity comes from antigen (DNA) binding site. Enzymatic constants and inhibitory constants showed that Ab G1-5 is one of the most efficient catalytic antibodies known so far.
