Flow field-flow fractionation (FlFFF) is an elution-based technique that is capable of separating and characterizing colloidal particles, proteins, and macromolecules according to the differences in hydrodynamic size of sample components. Xanthan is a branched, anionic microbial heteropolysaccharide produced by aerobic fermentation of the bacterium Xanthomonas campestris. Xanthan gum exist to main chain ?ß-D-glucose 1,4 bond and side chain?mannose, glucuronicacid mannose acetyl group, pyruvate group. Xanthan is widely and more extensively used as a food gum than is any polysaccharide (suspension, emulsifier, stabilizer, food applications, etc.) In this study FlFFF combined with multi angle light scattering (MALS) has been used to study the molar mass and molar mass distribution of xanthan gum and partially acid-hydrolyzed xanthan. A food grade xanthan sample was hydrolyzed in 0.1 N HCI at 80°C. Aliquots were drawn from the reaction bath, and the reaction stopped by quenching the samples to room temperature and neutralized to pH 6-7 by adding aqueous NaOH. Results show FlFFF-MALLS may play an important role in characterization of Xanthan gum particles for its applications for food by providing some additional informations on the sample.
