Spore is basidiospore. A single mushroom may produce up to 4 million spore per hour. Usually spores are of 8-10.5ⅹ 3-3.5 ? in size. They have smooth surface and cylindrical to narrowly kidney-shaped. Gravitational field-flow fractionation (GrFFF), is a separation technique that utilizes earth‘s gravity as the external field. GrFFF is a convenient and easy-to-operate technique that has shown to be suitable for the size and/or density-based separation of organic or inorganic micron-sized particles of various origins. In this study, GrFFF was used to separate the Oyster mushroom spores based on their sizes, which yields the size distribution of the spores. Then the slices of the GrFFF fractogram were analyzed by optical microscopy (OM) for determination of the size and the shape. The flow rate was varied from 0.15 to 0.2 mL/min for GrFFF optimization. Fractions were collected and then cultured to examine the dependence on the size. The results from GrFFF and OM indicated that the size and shape of the spores were varied with the different culturing time due to aggregation. Oyster mushroom spore was obtained using the high quality seed.
