Colorectal cancer is one of the most commonly diagnosed cancers in South Korea. Inherited deficiencies of polymorphisms in DNA repair genes may alter DNA repair efficiency and contribute to susceptibility to cancers. X-ray repair cross complementing group1 (XRCC1) is one of important DNA repair genes in base excision repair (BER) and interacts with several proteins, poly (ADP-ribose) polymerase (PARP), polymerse β, DNA ligaseⅢ, polynucleotide kinase (PNK) and AP endonucleaseⅠ. There are some reports that three codons of XRCC1, Arg194Trp, Arg280His and Arg399Gln, are associated with colorectal cancer. In this study, We investigated these three polymorphisms of XRCC1 in 44 colorectal cancer tissues, amplified polymorphic sites by PCR followed by digestion of restriction enzymes (MspⅠand RsaⅠ) and detected by denaturing high performance liquid chromatography (DHPLC). As a conclusion, our data show that Arg194Trp and Arg399Gln variants of XRCC1 may be associated with increasing risk of colorectal cancer with high frequencies.
