Identification of genome-wide target genes of transcription factors (TFs) should provide the biological significance of TFs but also the global transcriptional regulatory mechanism. We are using both siRNA and DNA microarray technologies to identify genome-wide target genes of human transcription factors. In this approach, TF-specific siRNA perturbs the expression of the TF’s target genes, and as a consequence, global transcription change is induced. The resulting change in transcriptome is analyzed using the DNA microarray. We are currently analyzing the transcriptome of individual human TFs such as NRSF and DREAM, as well as those of TF families, such as basic leucine zipper transcription factors. This approach will not only provide considerable information in identifying the novel targets of the TF, but also help in understanding the transcriptional regulatory network in human cells.
