One of the fundamental quests in synthetic biology is to construct designer genes and genomes in a high-throughput manner to probe and engineer biological systems. However, typically employed DNA synthesis procedures impose significant challenges for scalable construction due to the high cost of oligonucleotides, low assembly efficiency, requirement for time-consuming cloning and the high cost of sequence validation. In contrast to the slow progress in DNA synthesis, DNA sequencing technology has progressed rapidly in recent years to the level of de novo human genome sequencing. Here, I present recent progress on high-throughput DNA writing methods that are based on the combination of microchip oligonucleotides and high-throughput sequencing technology. Considering the fact that current high-throughput DNA sequencing technology can generate a few billion reads, and that microchip DNA density is approaching a few million spots, I envision that our synthetic methods will be further improved and will benefit genome writing and synthetic biology projects that require a large number of highly variable DNA molecules.
