Ganglioside GM1 is the natural receptor for cholera toxin (CT) and heat-labile enterotoxin (LT), which are the causative agents of cholera and traveler's diarrhea. The small molecules interfering with this recognition process may prevent entry of the toxins into intestinal cells, thereby averting their devastating effects. The terminal sugar of ganglioside GM1, galactose, has been chosen as a possible lead structure in designing such receptor antagonists. There have been investigated on designing novel ligands through sugar mimic approach, such as design of multivalent ligands or structure based modification of m-nitrophenyl-galactoside (MNPG). Natural pentasaccharides showed hydrogen bonding with Glu11, His13, Glu 51, Gln 61 and Asn90 according to the analysis of binding mode of crystal structure. In this study, de novo design approach was carried out for designing novel ligands that can show additional hydrogen bonds with His13 by attaching several side chains on the MNPG.
