Understanding the molecular mechanisms of synaptic vesicle fusion mediated by soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) is essential for studying neuronal communication. Although various in vitro single-vesicle assays monitoring the fusion process were developed before, they have several limitations for detecting the process of SNARE-mediated content mixing and dynamics of fusion pore, transient intermediated steps of vesicle fusion,. Here, we introduce a new, reliable and efficient single-vesicle content mixing assay using a high affinity, fluorophore tagged host-guest pair, cucurbit[7]uril-Cy3 and adamentane-Cy5 as a FRET pair. Using this systems, we can observe the series of conetnt mixing process from vesicle docking, initial fusion pore formation to complete fusion with a high signal to noise ratio. Especially, we successfully observe the reversable flickering of the fusion pore for the first time in vitro. These results demonstrate that the single vesicle content-mixing assay based on our new synthetic host-guest FRET pair is a powerful tool to study the detailed vesicle fusion processes.
