Hepatitis C virus (HCV) infection is global health problems that is cause of chronic liver disease worldwide with more than 185 million people at risk of developing liver cirrhosis and hepatocellular carcinoma. Since HCV displays a large degree of genomic variability, resulting in its quasispecies distribution. Variants that confer resistance to previous NS5A inhibitors pre-exist within HCV quasispecies populations. The virus-encoded nonstructural protein 5A (NS5A) is a multifunctional protein essential for HCV replication. Therefore, a number of potent lead compounds were synthesized for targeting NS5A and evaluated their activity against HCV, in which BMS-790052 as a reference compound. Among these compounds, several compounds showed potent anti-proliferative activity against HCV cell lines, less than 10 pM EC50. They could be new potential drugs for anti-HCV, and we continue to development new compounds for targeting NS5A.
