Lanthanide-doped upconversion nanoparticles (UCNPs) possess unique upconversion luminescence features, an excellent photo-stability compared with conventional bioimaging fluorophores and are composed of materials that are not particularly toxic or have tolerable toxicity to biological systems. Thus, UCNPs are considered to be very promising NIR bioimaging agents with the characteristics of high signal-to-noise contrast, high penetration depth and minimum photo-damage to biological system. UCNPS were synthesized by hydrothermal method using as template Melamine-Formaldehyde using YVO4:Yb3+, Eu3+, Er3+. Functionalization of UCNP by GSH (Glutathione) and APTES was attended by simple reaction in reflux. To investigate feasible applications in bioimaging, HeLa cells have been cultivated in DMEM (modified Eagle’s medium) supplemented with 10% FBS (fetal bovine serum) at 37℃ and 5% CO2. Cells (5ⅹ108/L) were plated on 14 mm glass coverslips and allowed to adhere for 24h.1 Subsequently, after washing with phosphate buffer solution (PBS), the cells were incubated in DMEM containing 0.3mg/mL of UCNP-GSH or UCNP-APTES and incubate for 1h, in 5% CO2. HeLa cells treated with Functionalized UCNP have been wrapped by UCNP after 1h. Cytotoxicity test showed that nanoparticles with APTES are toxic, with 60% cell viability. Further tests have confirmed that APTES molecules are basically toxic for biological systems. UCP-GSH nanoparticles were also toxic for HeLa cells despite GSH molecules are non-toxic for biological systems. Keywords: Bioimaging, UCNP, HeLa cells, Glutathione, APTES, Cytotoxicity
