Some heavy metal ions contaminated in ground water showed toxicity to human body. As(III) and Cu(II), among those metal ion, accumulated in human body and cause many severe diseases. We designed a fluorescent chemosensor based on ArsR regulator peptide sequence in human body for detecting heavy metal ions. The regulator contains cysteine as key amino acid and cysteine was well known for binding with copper. We observed the sensor bound with arsenic and some other metals, but after we treated EDTA, the metal chelator, it showed selective response to As(III) and Cu(II) among 17 metal ions. The binding mode of this sensor maybe the key factor. And we used pyrene fluorophore for ratiometric sensing so we can do quantitative analysis. Interestingly, when the sensor made dimer by forming disulfide bond via cysteine, metal selectivity of this sensor was changed. So, we have a plan to further studies about this.
