Matrix-Assisted Laser Desorption/Ionization (MALDI) is an ionization technique. This technique is usually used in analysis of macromolecules such as proteins. In MALDI, the most commonly used sample preparation method is dried droplet. In short, this method is composed of three steps; mixing sample/matrix, loading the mixture droplet on the MALDI plate and drying the droplet in room temperature or vacuum condition. Because of induced inhomogeneity of matrix crystal after drying procedure, we pay attention to get a homogeneous product in crystallization method. One of the ideas that we designed is freeze vacuum drying. In this new method, the sample/matrix mixture was first frozen rapidly at very low temperature. After freeze process, the solvent of frozen mixture was sublimated in low pressure by vacuum pump. We will show the accomplished homogeneous dried spot product and the mass spectra. A comparison to air drying and vacuum drying will also be shown.
