Therapeutic glycoproteins such as mAbs, growth factor, and enzymes have extremely heterogeneous glycosylation including isomeric glycans and modified glycans which influence on immune response, efficacy, and safety of drug. Porous graphitic carbon (PGC) based chromatography with mass spectrometry has been widely used to separate native (underivatized) glycan isomers, thus allowing simpler sample preparation. However, native glycan analysis often occurs anomer separation at the reducing end, thus by increasing complexity. Here, we described an efficient analytical platform to separate glycan isomers using PGC-based chromatography and to characterize glycan structure using tandem MS. To prevent an anomer separation, native glycan was reduced by NaBH4 prior to LC/MS analysis. Each peak having different retention time in a chromatogram was further isolated and elucidated using MS/MS. We applied this analytical platform to glycans released from therapeutic mAbs. We successfully separated glycans without anomer interference and identified various structural or linkage isomers.
