Identification of proteins related with estrogens-induced proliferation of human MCF7 breast cancer cells

17beta-estradiol (E2) and its metabolites such as 2-hydroxyestradiol, 4-hydroxyestradiol, 16alpha-hydroxyestrone and 2-methoxyestradiol are known to play an important role in the development and progression of breast cancer. To understand the functional role of E2 and its metabolites in breast cancers, we applied the proteomics technologies to identify proteins related with the E2 metabolites-induced proliferation of MCF-7 cells. When total 1087 proteins were analyzed, 29 proteins were > 2-fold increased and 16 proteins were > 2-fold decreased by E2 and/or its metabolites. These proteins were included in a variety of biological processes such as cytoskeleton organization, protein folding, protein phosphorylation, and reactive oxygen species metabolism. This study could be helpful to understand the mechanism to induce the proliferation of MCF-7 by estrogen metabolites.
