Water-soluble quantum dots (QDs) have been widely used in immunoassays, in situ hybridization and imaging studies. In biology they act as an excellent donor in fluorescence resonance energy transfer (FRET) based applications due to their strong photoluminescence narrow emission and broad excitation spectra, enabling the effective separation of the donor and acceptor fluorescence. In this study, we synthesized DHLA capped CdSe/ZnS QDs, and conjugated with Cy5-labelled or unlabelled oligonucleotide using EDC/NHS chemistry. As-synthesized oligonucleotide-QD conjugates were subjected to hybridize with the complementary unlabelled or Cy5-labelled target DNA. The hybridization reaction was monitored by static fluorospectroscopy using FRET from the QD to the Cy5. Treatment of the Cy5-labelled QD with EcoR1 resulted in the cleavage of the hybridized DNA and the recovery of the QDs’ fluorescence.
