The global effect of MicroRNAs on the protein expression in Arabidopsis thaliana and the expression changes of CSDPs and GRPs in E. coli and Arabidopsis thaliana under cold stress analyzed by 2DE/MS

MicroRNAs (miRNAs) are non-coding, approximately 20 nucleotide small RNAs. MiRNAs are not translated into proteins but show diverse and important functions in plant metabolism. The effect of miRNA over-expression on the expressional changes in Arabidopsis thaliana was examined by two-dimensional gel electrophoresis and MALDI-TOF MS. That is, the protein spots showing significant changes were identified by in-gel digestion and peptide fingerprinting with a MALDI-TOF mass spectrometry. Cold shock domain proteins (CSDPs) and glycine-rich RNA-binding proteins (GRPs) have influence on cold adaptation process in prokaryotes. To study the functional roles of plant CSDPs and GRPs in cold response, CSDPs and GRPs from Arabidopsis thaliana were investigated for their growth-stimulating abilities under cold stress of Escherichia coli BX04 mutant. This study revealed that several genes were up or down regulated by the expression of miRNAs, CSDP1 or GRP7 in E. coli and Arabidopsis thaliana.
