Rat brain protein identified by Ca2+-calmodulin affinity chromatography and MALDI-TOF mass spectrometry

Calmodulin, a 17 kDa protein with four calcium-binding sites, serves as a calcium sensor in nearly all eukaryotic cells. Calmodulin plays an important role in various signal transduction pathways through binding to a variety of target proteins. Ca2+-calmodulin-binding proteins from rat brain were harvested by an affinity chromatography using calmodulin-immobilized Sepharose. Subsequently, Calmodulin-binding proteins were further separated by 2-dimensonal gel electrophoresis and identified by MALDI-TOF mass spectrometry. About 270 proteins were resolved on a 2D gel and the 50 major proteins were identified. The method described in this study is very prospective for screening new calmodulin-binding enzymes.
