RNAi-mediated VEGF gene silencing could be an alternative approach for tumor-anti-angiogenesis and tumor suppression. However, the use of siRNA in clinical purposes is still questioned due to the problems with rapid enzymatic degradation and poor intracellular uptake of siRNA. In this study, synthetic VEGF siRNA was conjugated to polyethylene glycol (PEG) via a disulfide linkage to address the obstacles of siRNA delivery. The conjugate could generate polyelectrolyte complex micelles by interacting with PEI. The disulfide linkage between PEG and siRNA could be cleaved to release the siRNA in the reductive intracellular environment. The efficiency of the polyelectrolyte complex micellar formulation was shown by evaluating anticancer property of VEGF siRNA in human prostate carcinoma.
