Electrochemical study of mono and multilayer cytochrome c immobilized conducting polymer modified electrodes

Mono- and multi-layers of cytochrome c (cyt c) were assembled on a poly-TTCA monolayer modified electrode. The polymer film was grown electrochemically from a TTCA monolayer that was prepared onto the electrode by the LB method. CVs of cyt c covalently attached on a monolayer showed a pair of well-defined and reversible peaks at about 228/168 mV (Epa/Epc) vs Ag/AgCl (sat’d KCl) electrode in a 0.2 M phosphate buffer solution (pH 7.0). The electrochemical properties of immobilized cyt c were confirmed by CV and QCM. In addition, the surface morphology and molecular size of the TTCA monolayer, poly-TTCA monolayer, and cyt c immobilized poly-TTCA monolayer on the HOPG electrode were observed by AFM. Heterogeneous electron transfer rate constant (ks) of cyt c on the poly-TTCA monolayer was determined to be 0.874 sec-1, in which the ks) value decreased for the multilayer cyt c. The current-voltage (I-V) characteristics of Au/poly-TTCA/cyt c/poly-TTCA/Au device have been studied for bioelectronics applications.
