We have been trying to find out new drug candidate compounds for Alzheimer's drugs based on the cholinergic theory. 9,10-Dihydro-9-oxa-10-phosphaphenanthrene-10-oxide (DOPO) is one of phosphinates and it is well known as a flame retardant. Since many organophosphorus compounds were applied as insecticides such as ethyl parathion, malathion, methyl parathion, chlorpyrifos, diazinon, dichlorvos, phosmet, fenitrothion, tetrachlorvinphos, azinphos methyl, pirimiphos-methyl, dimethoate, and phosalone and nerve agents such as tabun, sarin, soman, cyclosarin, and VX by inhibiting acetylcholinesterase, we synthesized DOPO derivatives by the click reaction between propargylated DOPO and substituted benzyl azide. In vitro cholinesterase [acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE)] inhibitory activities were evaluated by using DOPO derivatives. The IC50 value of compound 1 is 3.14±0.02μM which is more effective than galantamine (IC50 = 9.4±2.5μM against BuChE).
