Intracellular viscosity is an important physical parameter and Nitric oxide (NO) is an essential gas signaling molecule involved in many physiological processes. Irregular viscosity in the cell and the production of NO cause various diseases. However, fluorescent probes capable of detecting both viscosity and NO have not been studied. So, we designed and synthesized probe <1> for dual imaging of viscosity and exogenous and endogenous NO in living cells. Probe <1> has no fluorescence due to N-N single bond free rotation. When the viscosity increases, free rotation is prohibited, resulting in significant blue fluorescence. In addition, probe <1> and NO can react to form compound 2, which emits a pronounced green fluorescence. Moreover, probe <1> had low toxicity in living cells, fluorescence was observed according to the viscosity change in the blue channel, and changes in exogenous and endogenous NO concentrations were observed by imaging in the green channel. Most notably, we were able to study the relationship between viscosity and NO in living cells with probe <1> for the first time, which can help with many pathological basic studies.
