Recent development of super-resolution fluorescence microscopy allows the location of molecules to be determined with nanometer-scale spatial resolution. Moreover, correlative super-resolution microscopy, the integration of the high spatial resolution of super-resolution fluorescence microscopy with other imaging platforms, offers intriguing opportunities to probe multiple aspects of a given system. In particular, assembling spectral information, together with the super localized positions of the same molecules, enables single-molecule spectroscopy study. In this presentation, I will talk about our recent technology development and applications of correlative STORM and spectroscopy(spectral STORM). By dispersing the emission of many single molecules simultaneously in the wide field and implementing fluorescence on–off switching, the spectra of millions of individual molecules can be obtained within minutes. Using this approach, we studied the solvatochromism and ring-opening reaction of photochromism molecules. Our approach will open the door to the simultaneous nanoscale structural imaging and local sensing of surface and soft-matter systems inaccessible to current methods.
