Translocator protein (TSPO) is an 18 kDa protein, which was previously known as the peripheral benzodiazepine receptor (PBR) and found on the mitochondrial outer membrane in eukaryotes. TSPO was used as a clinical neuroimaging biomarker of neurodegenerative diseases and neuroinflammation which has increased exponentially for over 10 years. There are many studies aimed to develop novel radiotracers for TSPO positron emission tomography (PET) imaging and their use have now been described extensively in many neurodegenerative diseases and mental disorders. We are interested in the development of novel diagnosis agents for brain injury and neuroinflammation. Herein, we describe the synthesis of a series of novel fluorinated TSPO ligands bearing pyrazolo[1,5-a]pyrimidin-3-yl, branch propoxyl, and dimethyl moieties. In additions, the most potential ligand precursor was radiolabeled with F-18 from the precursor to be applied for PET study.
