Urea is an important platform for organic compounds synthesis. Urea derivatives were also found in many biologically active compounds. Due to its importance in various fields, several synthetic procedures have been reported for formation of urea structures. However, they have some drawbacks such as utilization of highly toxic phosgene regent. The direct preparation of asymmetrical ureas from N-Cbz-protected amines was a challenging approach. In our investigations, we screened commercial available reagents such alkali metals and alkaline earth metals as catalysts to prepare asymmetrical ureas. The result showed that CaI2 was a useful catalyst to prepare target asymmetrical ureas. With the optimized conditions, a wide range of asymmetrical ureas were synthesized. Our results indicated that the novel CaI2-catalyzed direct conversion of N-Cbz-protected amines into asymmetrical ureas is applicable for the preparation of various useful ureas.
