In this study, we have developed correlative and quantitative super-resolution imaging technique to investigate the platelet activation mechanism. Correlative super-resolution microscopy, the integration of super-resolution fluorescence microscopy with other microscopy techniques performed on the same sample, produces results that emphasize the strengths of each technique while offsetting their individual weaknesses. Using this approach, we investigate the ultrastructural change of platelets in nanoscale resolution during its activation process by observing cytoskeletal elements, which has not been resolved with conventional microscopy. Our data present the detailed interaction between the different cytoskeletal elements during the platelet activation process. Further correlative imaging is expected to provide a better understanding of the platelet activation mechanism by identifying the detailed interaction between the cytoskeletal elements and other organelles during the activation process.
