Nucleosides have been long regarded as useful templates because they are not only DNA/RNA building blocks but also acting as cellular signaling agents. Several nucleoside analogues have been used in clinical as anticancer and antiviral agents in the result of the development of a variety of sugar modified nucleoside analogues. On the basis of bioisosteric rationale, the corresponding 4’-thio and 4’-methylene nucleosides were reported as biologically active compounds. We developed novel 4’-selenonucleoside analogues which include selenium as a bioisostere of oxygen, sulfur and methylene. From the x-ray crystallographic study, 4’-Selenonucleosides were discovered to adopt the C2’-endo/C3’-exo (south) conformation, which is different from that of oxo-nucleosides. Based on this conformational feature, novel 4’-selenonucleosides analogues have been designed, synthesized and evaluated anticancer and antiviral activities, and some analogues were developed A3 adenosine receptor agonists.
