3,N4-ethenocytosine (εC) is a exocyclic DNA base adduct produced either by exposure of cellular DNA to industrial carcinogen, vinyl chloride, or as a consequence of cellular lipid peroxidation. Double-strand uracil-DNA glycosylase (Dug) has been identified to play a dual role in excision of uracil (U) and εC. Comparative analysis of the substrate specificity revealed that Dug was considerably more active on εC-DNA relative to U-DNA. εC excision activity of Dug processes more than a stoichiometric amount of substrate, unlike U excision activity. Removal of both U and εC resulted in AP-site in DNA while Dug binds tightly to this DNA product. Analysis of the binding isotherm of Dug on AP-site DNA by evanescent wave biosensor determined Kd=2.38 nM for Dug on AP/G-DNA. Complete DNA repair of εC occurred solely dependent on the presence of Dug, suggesting important cellular role of Dug in mutation avoidance against chemical induced genotoxicity.
