The incorporation of reduced amide bond ψ[CH2NH] into peptides results in the increase of the net positive charge as well as the perturbation of a helical structure. By using this characteristic of reduced amide bond, we designed and synthesized novel pseudopeptides containing reduced amide bonds and investigated their binding affinities and leakage activities for large unilamellar vesicles at different pH. The structure-activity relationship study about pseudopeptides indicated that the decrease of a helicity as well as the increase of net positive charge in the backbone, caused by an incorporation of reduced amide bond into the peptide, play an important role in the binding activities and leakage activities.
