The goal of this study is to investigate kinetics of protein refolding along with ATP hydrolysis assisted by E. coli chaperone GroEL and folding modulators under permissive and non-permissive conditions.  In vitro protein refolding was assayed as followings with chaperone GroEL with or without GroES; (1) Real time kinetics of substrate protein refolding assisted by the purified GroEL chaperone system was monitored, (2) Effects of temperatures on the process of protein refolding were investigated, and (3) Correlation between the kinetics of protein refolding and the kinetics of ATP hydrolysis was studied.  We have also investigated effect of the presence of folding modulator such as DnaK chaperone on the rate of protein refolding and the yield of refolded protein will be investigated in order to test the hypothesis concerning the respective roles of the GroEL/GroES chaperone and the DnaK chaperone during the process of protein refolding.
